目的研究携抗基质金属蛋白酶-9抗体脂质微泡(TMB-9)对睾丸缺血再灌注的造影评价。方法 54只大白兔随机分成对照组(S组)、轻度组(R组:R1、R2、R3、R4组)、中度组(R′组:R′1、R′2、R′3、R′4组)。术前、术后每组分别进行睾丸脂质微泡(MB)及TMB-9超声造影。术后行睾丸免疫组织化学检测基质金属蛋白酶(MMP-9)。结果术后TMB-9组中,R1组及R′1组较S组PI、Slope、Area增加,DT/2延长,TP缩短(P<0.05)。随着复灌注时间延长,PI、Slope、Area降低,TP延长(P<0.05)。R组及R′组中,TMB-9组均较MB组PI、MTT增加,DT/2、Slope显著增加,TP降低(P<0.05)。随着复灌注时间延长,睾丸微血管基底膜MMP-9表达增多(P<0.05)。结论同一缺血复灌注条件下,TMB-9较MB能更好地评价睾丸微血管损伤程度。
Objective To study the contrast of testis ischemia-reperfusion with anti-MMP-9 lipid microbubbles (TMB-9). Methods Fifty - four rabbits were randomly divided into control group (S group), mild group (R group: R1, R2, R3 and R4 groups), moderate group (R ’group: R’1, R’2 and R’3 , R’4 group). Testicular lipid microbubbles (MB) and TMB-9 ultrasonography were performed preoperatively and postoperatively in each group. Postoperative testicular immunohistochemical detection of matrix metalloproteinase (MMP-9). Results In TMB-9 group, PI, Slope, Area increased, DT / 2 prolonged and TP decreased (P <0.05) in group R1 and group R’1. With the prolonged perfusion time, PI, Slope, Area decreased and TP prolonged (P <0.05). In R group and R ’group, PI and MTT in TMB-9 group increased, DT / 2 and Slope increased significantly and TP decreased (P <0.05) compared with MB group. With the prolonged perfusion time, the expression of MMP-9 in testis microvascular basement membrane increased (P <0.05). Conclusions TMB-9 is better than MB in evaluating the extent of testicular microvascular damage under the same conditions of ischemia reperfusion.
