目的研究14-3-3σ基因在鼻咽癌细胞株HNE1和耐顺铂鼻咽癌细胞株HNE1-DDP的表达情况,以了解14-3-3σ基因与耐药的关系。方法采用蛋白印迹法(western blotting,WB)检测HNE1-DDP细胞与HNE1细胞14-3-3σ的表达,并采用荧光定量RT-PCR(quantitative RT-PCR,q RT-PCR)检测14-3-3σ基因的mRNA表达水平。结果 14-3-3σ蛋白和其mRNA在HNE1-DDP细胞株中表达明显下降。结论 14-3-3σ下调可能与鼻咽癌耐药相关,其在耐药细胞株中表达下调的机制有待进一步的研究。
Objective To investigate the expression of 14-3-3σ gene in nasopharyngeal carcinoma cell line HNE1 and cisplatin-resistant nasopharyngeal carcinoma cell line HNE1-DDP in order to understand the relationship between 14-3-3σ gene and drug resistance. Methods Western blotting (WB) was used to detect the expression of 14-3-3σ in HNE1-DDP cells and HNE1 cells. The expression of 14-3-3σ was detected by quantitative RT-PCR (q RT-PCR) 3σ gene mRNA expression levels. Results The expression of 14-3-3σ protein and its mRNA were significantly decreased in HNE1-DDP cell line. Conclusion Down-regulation of 14-3-3σ may be related to drug resistance of NPC, and its mechanism of down-regulation in drug-resistant cell lines remains to be further studied.
