坤泰胶囊对卵泡凋亡相关蛋白Bcl-2/Bax的影响

目的:观察坤泰胶囊对卵泡凋亡相关蛋白Bcl-2/Bax的影响。方法:建立卵巢早衰小鼠模型,造模成功后随机分为正常组、模型组、戊酸雌二醇片组、坤泰胶囊组。正常组、模型组给予生理盐水,戊酸雌二醇片组和坤泰胶囊组均以0.2 mL/10 g的体积灌胃给予相应药物。实验末,取卵巢、子宫称重,计算脏器指数,并以放免法和免疫组化法分别检测小鼠血清E2、FSH的水平和卵巢组织Bax、Bcl-2蛋白的表达。结果:坤泰胶囊组小鼠卵巢质量比模型组明显增加,卵巢指数与模型组相比差异具有显著性(P<0.05);坤泰胶囊组小鼠血清中E2水平比模型组小鼠明显升高、FSH水平明显降低,相对模型组差异具有显著性(P<0.05);坤泰胶囊组小鼠卵巢组织Bax蛋白表达比模型组明显降低,Bcl-2蛋白表达比模型组小鼠明显升高,与模型组比较差异均具有显著性(P<0.05)。结论:坤泰胶囊可以促进卵巢的生长,并通过调节卵泡凋亡相关蛋白Bcl-2、Bax的表达来抑制卵巢功能早衰。 Objective: To observe the effect of Kuntai capsule on apoptosis-related protein Bcl-2 / Bax in follicle. Methods: The mouse model of premature ovarian failure was established. After the model was successfully established, the mice were randomly divided into normal group, model group, estradiol valerate group and Kun Tai capsule group. Normal group, model group given saline, estradiol valerate tablets group and kuntai capsule were given 0.2 mL / 10 g volume of the corresponding administration by gavage. At the end of the experiment, the ovaries and uterus were weighed and the organ index was calculated. The levels of serum E2 and FSH and the expressions of Bax and Bcl-2 protein in ovarian tissues were detected by radioimmunoassay and immunohistochemistry. Results: The ovary mass of mice in KunTai Capsule group was significantly higher than that in model group, and the difference of ovarian index was significant compared with model group (P <0.05). The serum E2 level in KunTai Capsule group was significantly higher than that in model group (P <0.05). The expression of Bax protein in ovary tissue of Kun Tai Capsule group was significantly lower than that of model group, while the expression of Bcl-2 protein was significantly higher than that of model group , Compared with the model group, the difference was significant (P <0.05). Conclusion: Kuntai capsule can promote ovarian growth and inhibit premature ovarian failure by regulating the expression of apoptosis-related proteins Bcl-2 and Bax.
