目的 分析纤维蛋白原(Fg)β-148 C/T基因多态性与性别、年龄、吸烟等因素对血浆Fg水平的影响。方法用PCR-RFLP的方法分析基因多态性,比浊法测定血浆Fg水平。结果病例组血浆Fg7k平明显高于对照组(P<.01),两组的T-148基因频率分布差异无显著性(P>0.05)。男女两性的血浆Fg水平比较差异无显著性(P>0.05):两性中T-148基因携带者血浆Fg水平均比 CC基因型者明显升高(P<0.05)。在 T-148携带者中,血浆 Fg 水平随年龄增长而升高 (P<0.05);吸烟组 Fg 水平高于不吸烟与戒烟组(P<0.05)。结论 Fgβ-148 C/T基因多态性与血浆 Fg水平相关,T-148基因携带者更易受年龄、吸烟等因素影响使血浆Fg水平升高。提示Fgβ-148 C/T基因多态性可作为血栓性疾病的遗传易感标志之一。
Objective To analyze the effect of fibrinogen (Fg) β-148 C / T gene polymorphism on plasma Fg levels, such as sex, age and smoking. Methods The gene polymorphism was analyzed by PCR-RFLP and plasma Fg level by turbidimetry. Results The plasma level of Fg7k in the patients was significantly higher than that in the control group (P <.01). There was no significant difference in the frequency of T-148 gene between the two groups (P> 0.05). There was no significant difference in plasma Fg levels between men and women (P> 0.05). Plasma Fg levels of T-148 carriers in both genders were significantly higher than those in CC genotypes (P <0.05). In T-148 carriers, plasma Fg level increased with age (P <0.05), while Fg level in smoking group was higher than that in non-smoking group and smoking cessation group (P <0.05). Conclusion Fgβ-148 C / T gene polymorphism is correlated with plasma Fg level. T-148 gene carriers are more likely to be affected by age, smoking and other factors to increase plasma Fg levels. It is suggested that Fgβ-148 C / T gene polymorphism may be one of the genetic susceptible markers of thrombotic diseases.
