目的:探讨顺铂对卵巢颗粒细胞的影响及二仙汤含药血清改善大鼠卵巢颗粒细胞损伤的作用机制.方法:选取大鼠原代卵巢颗粒细胞体外培养及顺铂处理的方法,将顺铂处理后卵巢颗粒细胞随机分为:cDDP组、cDDP+雌二醇组、cD-DP+二仙汤组,另设正常组,加入相应药理血清连续培养48 h.采用Western Blotting法检测各组大鼠卵巢颗粒细胞bcl-2、bax蛋白表达;RT-PCR法检测各组大鼠卵巢颗粒细胞bcl-2、bax mRNA表达.结果:与正常对照组比较,cDDP组bax mRNA及蛋白表达上调,差异有统计学意义(P<0.05).与cDDP组比较,CDDP+雌二醇组及cDDP+二仙汤组Bax mR-NA及蛋白表达均下调,差异有统计学意义(P<0.05).与正常组比较,cDDP组bcl-2 mRNA水平及蛋白表达下调,差异有统计学意义(P<0.05).与cDDP组比较,CDDP+雌二醇组及cDDP+二仙汤组bcl-2 mRNA转录及蛋白表达上调,差异有统计学意义(P<0.05).结论:二仙汤能够抑制cDDP诱导的卵巢颗粒细胞损伤,其作用机制可能与调节bcl-2/bax平衡有关.“,”Objective To explore the effects of cisplatin on ovarian granulose cells and the mechanism of Erxian Decoction containing serum improving the damage of ovarian granulose cells in rats. Methods: The cisplatin-treated cells by in vitro culture and cisplatin treatment of rat primary ovarian granulose cells were randomly divided into a CDDP group, CDDP + estradiol group and CDDP + Erxian Decoction group. Normal serum group was added to the corresponding pharmacological serum for 48 h. The expression levels of Bcl-2 and Bax in ovarian granulose cells of rats in each group were detected by Western Blotting method,and the expression levels of Bcl-2 and Bax in Ovarian granulose cells of rats in each group were detected by RT-PCR. Results: Compared with the normal control group, the expression level of Bax gene and protein in CDDP group was up-regulated, with significant difference (P<0.05 or P<0.01). Compared with CDDP group, the expression of Bax gene and protein in CDDP + estradiol group and CDDP + Erxian Decoction group decreased significantly (P<0.05).Compared with the normal group, the expression of Bcl-2 in CDDP group decreased significantly (P<0.05 or P<0.01). Compared with CDDP group, the transcription level of bcl-2 gene in CDDP + estradiol group and CDDP + Erxian Decoction group increased significantly (P<0.05). Conclusion: Erxian Decoction can effectively inhibit the damage of ovarian granulose cells induced by CDDP, and its mechanism may be related to the regulation of bcl-2/bax balance.
