目的:探讨丝裂霉素对胃癌细胞谷胱甘肽转移酶(GlutathioneS-transferase,GSTπ)表达的影响。方法:利用人胃癌细胞株BGC-823进行培养,加入丝裂霉素后5h和10h后采集细胞,进行免疫组化染色。结果:5h组,胞质中GSTπ含量下降,胞核中GSTπ增多;10h组其胞质与胞核中仍有少量GSTπ表达。结论:丝裂霉素对BGC-823癌细胞有明显的杀伤作用,其途径之一可能与抑制GSTπ基因表达有关。
Objective: To investigate the effect of mitomycin on the expression of glutathione S-transferase (GSTπ) in gastric cancer cells. Methods: The human gastric cancer cell line BGC-823 was used for culture. After adding mitomycin, the cells were harvested at 5h and 10h, and then immunohistochemically stained. Results: In the 5h group, the content of GSTπ in the cytoplasm decreased and the GSTπ in the nucleus increased. In the 10h group, there was still a small amount of GSTπ expression in the cytoplasm and nucleus. CONCLUSION: Mitomycin has obvious killing effect on BGC-823 cancer cells, and one of the pathways may be related to the inhibition of GSTπ gene expression.
