目的研究文昌鱼多糖(Polysaccharide fromBranchiostoma belcheri,PBB)对荷S180小鼠免疫功能的影响。方法 Horn法检测PBB在小鼠体内应用的安全性(LD50);建立S180荷瘤鼠模型,随机分组,实验组分别灌胃给予25、50、100mg.kg-1PBB,每日一次,持续2w。测定PBB对荷瘤鼠的抑瘤率、胸腺指数、脾指数的影响;全自动生化分析仪检测各组小鼠血浆免疫球蛋白IgA、IgG、IgM含量;放射免疫法检测各组小鼠血清IL-6和TNF-α水平。结果 PBB小鼠体内给予未见明显的中毒症状,无动物死亡,其LD50>3160mg.kg-1;给予上述PBB剂量2w后,抑瘤率分别达到39.1%、47.5%和53.4%;实验组IgA、IgG、和IgM含量、TNF-α和IL-6水平以及胸腺和脾指数均较模型组明显提高。结论文昌鱼多糖毒性低,使用安全,可明显抑制S180肉瘤的生长,其作用机制可能与PBB提高机体的免疫功能有关。
Objective To investigate the effect of Polysaccharide from Branchiostoma belcheri (PBB) on immune function in S180-bearing mice. Methods The Horn method was used to detect the safety of PBB in mice. The S180 tumor-bearing mice model was established and randomly divided into groups. The experimental groups were given gavage with 25, 50 and 100 mg.kg-1 PBB respectively, once daily for 2 weeks. The inhibitory rate of tumor, thymus index and spleen index of PBB in mice were measured. The contents of IgA, IgG and IgM in plasma of mice in each group were detected by automatic biochemical analyzer. The levels of serum IL -6 and TNF-α levels. Results PBB mice showed no obvious symptoms of poisoning and no animals died. The LD50 was 3160 mg.kg-1. After 2 weeks of administration of PBB, the tumor-inhibitory rates were 39.1%, 47.5% and 53.4% , IgG, IgM levels, TNF-α and IL-6 levels and thymus and spleen index were significantly higher than the model group. Conclusion The polysaccharides from amphioxus have low toxicity and safe use, which can significantly inhibit the growth of S180 sarcoma. The mechanism may be related to the enhancement of immune function by PBB.
