目的:研究金欣口服液对RSV感染大鼠体内Ⅰ型干扰素相关蛋白STAT1、STAT2和P-STAT1、P-STAT2表达的调控趋势。方法:将大鼠分为正常对照组(A组)、感染模型组(B组)、金欣口服液低剂量组(C组)、金欣口服液中剂量组(D组)、金欣口服液高剂量组(E组)、预防组(F组)、利巴韦林组(G组)7组,成功造模后,分别采用ELISA和RT-PCR检测各组大鼠肺组织内P-STAT1、P-STAT2和STAT1、STAT2的表达。结果:感染模型组较正常对照组、预防组和金欣口服液低剂量组未能上调大鼠体内P-STAT1和P-STAT2蛋白表达;而金欣口服液中、高剂量及利巴韦林组明显上调感染模型组RSV大鼠体内的P-STAT1和P-STAT2蛋白的表达(P<0.05,P<0.01)。结论:实验表明金欣口服液可通过上调P-STAT1和P-STAT2的表达,诱导IFN的产生,发挥抗病毒作用。
Objective: To study the regulation of Jinxin Oral Liquid on the expression of type I interferon-related protein STAT1, STAT2, P-STAT1 and P-STAT2 in rats with RSV infection. Methods: The rats were divided into normal control group (group A), infection model group (group B), Jinxin oral liquid low dose group (group C), Jinxin oral liquid medium dose group (group D), Jinxin oral High dose group (E group), prophylaxis group (F group) and ribavirin group (G group). After successfully modeling, the levels of P- STAT1, P-STAT2 and STAT1, STAT2 expression. Results: Compared with normal control group, prophylaxis group and Jinxin oral liquid low dose group, the expression of P-STAT1 and P-STAT2 protein in rats in model group was not up-regulated; while Jinxin oral liquid, high dose and ribavirin Group significantly up-regulated the expression of P-STAT1 and P-STAT2 protein in RSV-infected rats (P <0.05, P <0.01). Conclusion: The experiment shows that Jinxin Oral Liquid can induce the production of IFN by up-regulating the expressions of P-STAT1 and P-STAT2 and exert anti-virus effect.
