目的 :研究低血清培养条件下转化生长因子 β(TGF β)、胰岛素样生长因子 Ⅰ (IGF Ⅰ )、碱性成纤维生长因子(bFGF)对人髁突软骨细胞Ⅰ、Ⅱ、Ⅲ型前胶原基因表达的调控作用。方法 :采用体外细胞培养及mRNA狭缝杂交的方法。结果 :IGF Ⅰ对 3种前胶原的mRNA水平无显著影响。bFGF、TGF β均呈不同程度地抑制Ⅱ型胶原的基因表达 ,分别为对照组的 0 35 2及 0 6 5 8倍 ,同时TGF β显著增加了Ⅰ型胶原的基因表达。 结论 :IGF Ⅰ具有稳定软骨细胞表型的作用 ,bFGF、TGF β可抑制软骨细胞终末分化。
Objective: To investigate the effect of transforming growth factor β (TGFβ), insulin - like growth factor Ⅰ (IGF Ⅰ) and basic fibroblast growth factor (bFGF) on the expression of type Ⅰ, Ⅱ and Ⅲ procollagen in human condylar chondrocytes Regulation of gene expression. Methods: In vitro cell culture and mRNA slot hybridization method. Results: IGF Ⅰ had no significant effect on the mRNA levels of three procollagen. bFGF and TGFβ all inhibited gene expression of type Ⅱ collagen to varying degrees, which were respectively 0 35 2 and 0 658 times of the control group, meanwhile, TGF β significantly increased the gene expression of type Ⅰ collagen. Conclusion: IGF Ⅰ can stabilize the phenotype of chondrocytes, and bFGF and TGF β can inhibit the terminal differentiation of chondrocytes.
