采用改进的Elson-Morgan法及改良的咔唑法测定透明质酸中的氨基葡萄糖和葡萄糖醛酸的含量,筛选出氨基葡萄糖测定的最佳水解时间为6mol/L盐酸水解2h,回收率为 96.6%(n=6).改进后的Elson-Morgan法灵敏度提高2.25倍,重现性及线性良好,适用于透明质酸的定量分析.
The content of glucosamine and glucuronidase in hyaluronic acid was determined by modified Elson-Morgan method and modified carbazole method. The optimal hydrolysis time of glucosamine was 6mol / L hydrochloric acid hydrolysis for 2h, the recovery was 96.6 % (n = 6). The improved Elson-Morgan method has a sensitivity of 2.25 times higher reproducibility and linearity and is suitable for the quantitative analysis of hyaluronic acid.
