目的运用电感耦合等离子体质谱法(ICP-MS)直接测定儿童全血中Mg、Ca、Cu、Zn、Mn、Pb、Cd 7种元素的含量。方法优化了ICP-MS的工作参数,以6Li、45Sc、72Ge、115In、209Bi作为内标物质,儿童血液经稀释液0.1%Triton X-100和0.1%HNO3稀释20倍后直接用ICP-MS测定分析,用所建立的方法对232名幼儿园儿童血液样本进行检测分析。结果 7种元素标准曲线的线性关系良好,待测元素的相关系数为0.998 992~0.999 992。Mg、Ca、Cu、Zn、Mn、Pb、Cd方法的检出限分别为0.030 mg/L、0.740 mg/L、0.030 mg/L、0.007 mg/L、2.506μg/L、0.619μg/L、1.181μg/L,加标回收率为88.90%~120.00%,方法的精密度≤2.774%。此幼儿园儿童存在缺镁、缺铜、缺锌的现象,另外有2人出现镉超标,9人出现铅超标,且超标率分别为0.86%和3.88%。结论该方法灵敏、准确、稳定,可应用于全血中微量元素的检测分析。
Objective To directly determine the contents of seven elements of Mg, Ca, Cu, Zn, Mn, Pb and Cd in whole blood of children using inductively coupled plasma mass spectrometry (ICP-MS). Methods The working parameters of ICP-MS were optimized, and 6Li, 45Sc, 72Ge, 115In and 209Bi were used as internal standards. The blood was diluted 20 times with 0.1% Triton X-100 and 0.1% Analysis, with the established method of 232 kindergarten children’s blood samples for testing analysis. Results The calibration curve of seven kinds of elements had a good linearity. The correlation coefficient of the tested elements was 0.998 992 ~ 0.999 992. The detection limits of Mg, Ca, Cu, Zn, Mn, Pb and Cd were 0.030 mg / L, 0.740 mg / L, 0.030 mg / L, 0.007 mg / L, 1.181μg / L, the spiked recoveries were 88.90% ~ 120.00%, the precision of the method was less than 2.74%. The kindergarten children had magnesium deficiency, lack of copper and zinc deficiency. In addition, two people had excessive cadmium, nine people had excessive lead, and the exceeding standard rates were 0.86% and 3.88% respectively. Conclusion The method is sensitive, accurate and stable and can be applied to the detection and analysis of trace elements in whole blood.
