蝴蝶兰类原球茎诱导、增殖及植株再生条件初步研究

为优化蝴蝶兰类原球茎诱导和增殖再生条件,以蝴蝶兰品种‘红太阳’无菌播种形成的原球茎为材料,研究了取材时间、植物生长调节剂(6-BA、NAA)、碳源等对类原球茎诱导和增殖及分化再生的影响,结果表明,生长45d的蝴蝶兰原球茎最适于类原球茎诱导和增殖培养。最佳的类原球茎诱导和增殖培养基为:花宝1号3.5g/L+6-BA 5.0mg/L+NAA 0.7mg/L+白糖20g/L+琼脂6.0g/L+活性炭2.0g/L,pH为5.4-5.8,最高增殖系数可以达到17.33,比已报道的最高增殖系数提高了35.39%;类原球茎最佳分化培养基为:花宝1号3.5g/L+6-BA 3.0mg/L+NAA 0.5mg/L+白糖20g/L+琼脂6.0g/L+活性炭2.0g/L,pH为5.4-5.8,最高分化率可达96.67%,生根率最高可达到92%。 In order to optimize the conditions for the induction and proliferation regeneration of Phalaenopsis protocorms, the protocorm-like bodies formed by aseptic sowing of the Phalaenopsis species “Red Sun” were used as material to study the effects of time, plant growth regulators (6-BA, NAA) The results showed that Phalaenopsis protocorms grown 45 days were most suitable for the induction and proliferation of protocorm-like bodies. The best culture medium for protocorm-like induction and proliferation was as follows: Hua Bao 1 3.5g / L + 6-BA 5.0mg / L + NAA 0.7mg / L + sugar 20g / L + agar 6.0g / L + activated carbon 2.0g / The highest proliferation coefficient was 17.33, which was 35.39% higher than the highest reported proliferation coefficient. The optimum culture medium for protocorm-like bodies was: Hua Bao No.1 3.5g / L + 6-BA 3.0mg / L + NAA 0.5mg / L + sugar 20g / L + agar 6.0g / L + activated carbon 2.0g / L, pH 5.4-5.8, the highest differentiation rate of 96.67%, the highest rooting rate can reach 92%.
