目的 :寻找染色体 3 p14 .2区域与鼻咽癌相关的表达序列标记 (expressedsequencetag ,EST) ,为筛选鼻咽癌候选基因打下基础。方法 :运用生物信息学同源性比较筛选EST的策略 ,结合Northern杂交和逆转录PCR方法 ,检测 3 p14 .2区域的相关ESTs在鼻咽癌和正常鼻咽组织中的表达。结果 :在 3 p14 .2区域筛选到一个在鼻咽癌中低表达的ESTw2 3 3 12 ,与正常鼻咽上皮组织相比较 ,在鼻咽癌细胞株及 13例鼻咽癌活检组织中的 5例其表达下调 (P <0 .0 1)。结论 :染色体 3p14 .2区域ESTw2 3 3 12在鼻咽癌中表达下调 ,提示其可能参与鼻咽癌癌变过程 ,为克隆鼻咽癌相关基因提供了新线索
OBJECTIVE: To find expression sequence tags (ESTs) associated with nasopharyngeal carcinoma in the region of 3 p14.2 on chromosome 3 and lay a foundation for screening candidate genes of nasopharyngeal carcinoma. Methods: The ESTs of screening ESTs were compared using homology of bioinformatics. Northern blotting and RT-PCR were used to detect the expression of 3 p14 .2 region in nasopharyngeal carcinoma and normal nasopharyngeal tissues. Results: A low expression of ESTw2 3 3 12 in nasopharyngeal carcinoma was screened in the region of 3 p14.2. Compared with the normal nasopharyngeal epithelial tissue, 5 of nasopharyngeal carcinoma cell lines and 13 nasopharyngeal carcinoma biopsies Its expression was down-regulated (P <0.01). Conclusion: Down-regulation of ESTw2 3 3 12 in chromosome 3p14. 2 region in nasopharyngeal carcinoma suggests that it may be involved in the carcinogenesis of nasopharyngeal carcinoma and provides new clues for cloning NPC-related genes
