目的:研究制备抗皂角毒和表皮生长因子受体双功能抗体,用以导向皂角毒杀伤肿瘤细胞。方法:在制备EQ75、CY12.14单克隆抗体细胞株的基础上,通过杂交杂交瘤技术制备双功能抗体细胞株。结果:双功能抗体(EQ75×CY12.14)与单用皂角毒杀伤肿瘤细胞(A431)的IC50分别为10-9.6、10-8.4mol/L。结论:双功能抗体(EQ75×CY12.14)杀伤肿瘤细胞(A431),以IC50值计算,有效率为单用皂角毒的15倍左右。
OBJECTIVE: To prepare bifunctional antibodies against scutellariae gondii and epidermal growth factor receptors, which are used to direct cytotoxic effects of saponin. Methods: Based on the preparation of EQ75 and CY12.14 monoclonal antibody cell lines, bispecific antibody cell lines were prepared by hybrid hybridoma technology. RESULTS: The IC50 of bifunctional antibody (EQ75 x CY12.14) and killing of tumor cells (A431) with saponin alone was 10-9.6 and 10-8.4 mol/L, respectively. Conclusion: The dual function antibody (EQ75×CY12.14) kills the tumor cells (A431). The effective rate is calculated to be about 15 times that of saponin alone.
