目的研究日本血吸虫成虫可溶性抗原38kDa(SjAWA38kDa)的动物保护作用,并对SjAWA38kDa作为抗日本血吸虫病分子疫苗的可行性进行评估。方法用SDS-PAGE电泳、切胶、电洗脱、超滤等方法分离纯化SjAWA38kDa,继而用SjAWA38kDa免疫昆明小鼠,然后进行尾蚴攻击感染(40尾/只),进行减虫率和减卵率研究。结果SjAWA 38kDa天然蛋白质分子能刺激机体产生抗日本血吸虫的作用,其减虫率为33.8%,减卵率为51.7%,有统计学意义(P<0.05)。结论SjAWA 38kDa分子抗原具有刺激机体产生抗日本血吸虫病的免疫保护作用,可以作为抗日本血吸虫病分子疫苗研究的靶标。
Objective To study the protective effect of 38 kDa (SjAWA38kDa) soluble antigen of adult Schistosoma japonicum and evaluate the feasibility of SjAWA38kDa as a molecular vaccine against Japanese schistosomiasis. Methods SjAWA38kDa was isolated and purified by SDS-PAGE electrophoresis, gel electrophoresis, electroelution, ultrafiltration and so on. Then, Kunming mice were immunized with SjAWA38kDa and then cercariae challenge infection (40pcs / mouse) the study. Results SjAWA 38kDa natural protein could stimulate the body to produce anti-Schistosoma japonicum. The worm reduction rate was 33.8% and the oviposition rate was 51.7% (P <0.05). Conclusion SjAWA 38kDa antigen can stimulate the body to produce anti-schistosomiasis immune protection and can be used as a target for molecular vaccine against Schistosoma japonicum.
