人中黄的HPLC指纹图谱及炮制机理初探

目的:建立中药人中黄HPLC指纹图谱测定方法,为其质量评价提供参考。指认其中主要成分,并对生制品进行化学成分比较,探析人中黄的炮制机制,为创立新的科学制备方法奠定基础。方法:HPLC法建立中药人中黄指纹图谱,对照品确定有效成分,“中药色谱指纹图谱相似度评价系统(A版)”分析不同样品图谱的相似度,镜像图谱对比分析人中黄共有模式图与甘草指纹图谱的区别。结果:对12批市售人中黄药材进行相似度评价,标定了18个共有峰;市售人中黄与共有模式图谱的相似度为0.8~0.9。自制样品与共有模式图谱的相似度均小于0.9。标定出6种成分,5个共有色谱峰分别是甘草酸、甘草素、异甘草素、甘草查尔酮A和甘草次酸,1个非共有峰甘草苷。方法学考察结果符合指纹图谱要求,稳定良好。人中黄指纹图谱与甘草指纹图谱镜像对比结果证明人中黄化学成分发生较大变化,差异性明显。结论:建立了人中黄的HPLC指纹图谱,相似度高,精密度、稳定性、重复性均表现良好,揭示人中黄与甘草相比化学成分在炮制过程中的变化趋势,为人中黄药材质量控制标准提供参考。 OBJECTIVE: To establish a method for the HPLC fingerprint determination of Chinese traditional medicine Huang Zhong Huang for its quality evaluation. Results: The similarity of the yellow medicinal herbs in 12 batches of commercial people was evaluated and 18 common peaks were calibrated. The similarity of commercial yellow and common patterns was 0.8 ~ 0.9. The similarity between self-made samples and common mode spectra are less than 0.9. The six components were calibrated. The five common peaks were glycyrrhizin, glycyrrhizin, isoliquiritigenin, licochalcone A and glycyrrhetinic acid, and one non-shared peak glycyrrhizin. Methodological findings in line with the requirements of the fingerprints, stable and good. Comparison of human yellow fingerprinting and licorice fingerprinting mirrors shows that there is a great change in the chemical constituents of yellow, with obvious differences. Conclusion: HPLC fingerprint of human yellow was established. The similarity, precision, stability and reproducibility of the HPLC fingerprint of human yellow were well established. The change trend of the chemical components in the medium yellow and licorice compared with licorice was revealed. Quality control standards for reference.
