在心血管疾病的发生过程中,常出现内皮功能紊乱等初期症状,而此过程与一氧化氮(NO)介导的血管舒张密切相关,NO的释放是由内皮一氧化氮合酶NOS3所调控。近期研究表明,心血管疾病常常伴随着NOS3基因多态性变化及表观遗传学修饰,而现有研究对于NOS3表观遗传学及基因多态性对于心血管疾病调控机制及靶向治疗研究关注较少。本文综述了近年来关于NOS3在心血管系统相关疾病中的研究现状,总结了其在各类疾病发病过程中的分子机制,重点阐述了NOS3蛋白解离、NOS3表观遗传修饰及多态性在心血管疾病发生发展中的作用,并为相关疾病药物开发的治疗靶点设计提供参考。
