目的观察清热化湿、活血泻浊法对肾小球硬化大鼠肾组织病理的影响。方法将大鼠分为正常组、假手术组、模型组、尿毒清组、经验方组,后3组采用5/6肾切除法制作肾小球硬化大鼠模型,并给予相应药物灌胃4周,取肾组织切片,行HE、PAS染色,观察大鼠肾脏病理变化,计算肾小球硬化指数(GSI);采用数码医学图像分析系统求得基质阳性面积比值。结果与正常组及假手术组比较,模型组GSI、基质阳性面积比值均升高(P均<0.05)。与模型组比较,经验方组GSI、基质阳性面积比值有降低(P均<0.05);而尿毒清组差异无统计学意义(P>0.05)。结论清热化湿、活血泻浊组成的经验方能够改善模型大鼠肾脏病理,降低GSI及基质阳性面积比值,改善肾小球硬化的程度。
Objective To observe the effect of clearing heat, dampness, promoting blood circulation and removing turbidity on renal tissue pathology in rats with glomerulosclerosis. Methods The rats were divided into normal group, sham operation group, model group, niaoduqing group and experience square group. The rats in the latter three groups were made into model of glomerulosclerosis by 5/6 nephrectomy. The rats were given gavage 4 The kidneys were sectioned and stained with HE and PAS. The renal pathological changes were observed and the glomerular sclerosis index (GSI) was calculated. The ratio of positive area of matrix was calculated by digital medical image analysis system. Results Compared with the normal group and the sham operation group, the GSI and matrix positive area ratio in the model group were all increased (all P <0.05). Compared with the model group, the ratio of GSI and matrix positive area in the experimental group was lower than that in the model group (all P <0.05), while there was no significant difference between the two groups (P> 0.05). Conclusion The prescriptions of clearing heat and dampness and activating blood and diarrhea can improve renal pathology, reduce the ratio of GSI and matrix positive area, and improve the degree of glomerulosclerosis.
