目的:观察福尔马林致痛和针刺镇痛时大鼠脑内孤啡肽免疫阳性物质和孤啡肽mRNA的变化情况。方法:大鼠80只,分为4组:①生理盐水组,在动物右后脚掌皮下注射生理盐水;②电针组,在大鼠右后肢的“足三里”和“昆仑”2穴上给以电针;③福尔马林组,在动物右后脚掌皮下注射 5g/L福尔马林;④电针+福尔马林组,给大鼠电针第 10 min时,在同侧后脚掌皮下注射福尔马林。采用免疫组织化学和原位杂交技术观察大鼠脑内孤啡肽的变化。结果:福尔马林致痛和电针 30 min时脑内孤啡肽免疫阳性细胞数明显减少,电针后再给动物注射福尔马林,孤啡肽免疫阳性细胞数介于福尔马林致痛和电针镇痛组之间,而不是进一步减少。福尔马林致痛和电针10h后,脑内孤啡肽mRNA阳性细胞数增加,电针后再给动物注射福尔马林,孤啡肽mRNA的表达进一步增加。结论:福尔马林致痛和电针均可促进孤啡肽的释放及合成,激活内源性的孤啡肽能系统。提示电针镇痛时脑内孤啡肽的释放及合成的增加可能是针刺效果不全的原因之一。
Objective: To observe the change of orphanin immunopositive substance and orphanin peptide mRNA in rat brain induced by formalin and analgesia. Methods: Eighty rats were divided into four groups: ① saline group, injected subcutaneously with normal saline in the right posterior palmar; ② electroacupuncture group given “Zusanli” and “Kunlun” The rats in the formalin group were injected subcutaneously with 5 g / L formalin in the right posterior palmar of the animals. The rats in the EA and formalin groups were given ipsilateral 10 min after electroacupuncture Feet subcutaneous injection of formalin. Immunohistochemistry and in situ hybridization were used to observe the change of orphanin in the rat brain. Results: Formalin induced pain and electroacupuncture at 30 min significantly reduced the number of orphanin immunopositive cells in the brain. Animals were injected with formalin after electroacupuncture. The number of orphanin-immunopositive cells was between Formal Lin Zhi Tong and electroacupuncture analgesia group, rather than further reduction. After induced by formalin and electroacupuncture for 10h, the number of positive cells of orphanin peptide in the brain increased, and then the animals were injected with formalin after electroacupuncture. The expression of orphanin mRNA was further increased. CONCLUSION: Both formalin induced pain and electroacupuncture can promote the release and synthesis of orphanin and activate the endogenous orphaninopeptidase system. Prompt electroacupuncture analgesia brain release of orphanin peptide synthesis and the increase may be one of the reasons acupuncture ineffective.
