目的观察高浓度葡萄糖对雪旺细胞增殖及凋亡的影响并初步探讨其可能机制。方法将体外培养的RSC96雪旺细胞分为正常对照组(25 mmol/L)和高糖组(100 mmol/L),分别应用M TT及流式细胞技术观察高糖对RSC96细胞增殖及细胞凋亡的影响。应用Western blotting观察高糖对Lipin-1、神经生长因子(NGF)蛋白表达的影响,并应用免疫荧光技术进一步验证在高糖刺激下Lipin-1蛋白表达变化。结果与正常对照组比较,高糖组对RSC96细胞增殖具有显著抑制作用(P<0.05)。与正常对照组晚期凋亡率(1.56±0.72)%相比,高糖组细胞晚期凋亡率(12.40±0.65)%明显增加。高糖显著降低Lipin-1和NGF蛋白表达(P<0.05)。结论高糖显著抑制雪旺细胞增殖并促进细胞凋亡;同时显著降低了Lipin-1和NGF的蛋白表达。提示高糖对神经细胞的损伤作用可能存在多种机制。
Objective To observe the effect of high concentration glucose on proliferation and apoptosis of Schwann cells and to explore its possible mechanism. Methods Schwann cells cultured in vitro were divided into normal control group (25 mmol / L) and high glucose group (100 mmol / L). Compared with the late apoptosis rate (1.56 ± 0.72)% in the normal control group, the late apoptosis rate in the high glucose group (12.40 ± 0.65)% was significantly increased. High glucose significantly decreased Lipin-1 and NGF protein expression (P <0.05). Conclusion High glucose can significantly inhibit the proliferation of Schwann cells and promote apoptosis. Meanwhile, the protein expression of Lipin-1 and NGF was significantly decreased. Tip high glucose on nerve cell damage may exist a variety of mechanisms.
