目的:建立一种从肉桂中同时分离5种苯丙素类化合物的制备分析方法,并对化合物的免疫抑制活性进行了评价。方法:肉桂经乙醇提取后,取乙酸乙酯部位经高效液相色谱仪分离制备化合物,制备色谱条件:采用Ultimate XB-C18(250 mm×10 mm,5μm)半制备柱,流动相为乙腈-水,进行梯度洗脱,检测波长为330 nm,柱温为30℃,流速为2.5 ml·min-1,进样量为0.3 ml。采用MS及NMR对化合物进行结构鉴定。采用CCK-8法对分离出的化合物进行免疫抑制活性考察。结果:同时分离得到5种苯丙素类化合物,分别为赤式-愈创木甘油、(7R,8S)-syringoylglycerol、(7S,8S)-syringoylglycerol、3-甲氧基苯基-丙烯醛、邻甲氧基肉桂醛。CCK-8法证明化合物3在浓度800μmol·L-1时对T细胞及B细胞的抑制率均大于20%,有一定免疫抑制作用。结论:苯丙素分离方法简便,分离效果好,能同时制备肉桂中5种苯丙素类化合物,其中化合物3有一定的免疫抑制活性。
OBJECTIVE: To establish a method for the simultaneous preparation and separation of five phenylpropanoids from cinnamon and to evaluate the immunosuppressive activity of the compounds. Methods: After the cinnamon was extracted by ethanol, the ethyl acetate fraction was separated by high performance liquid chromatography to prepare the compound. Preparative column chromatography was performed on a semi-preparative column of Ultimate XB-C18 (250 mm × 10 mm, 5 μm) with mobile phase of acetonitrile- Water gradient elution at a detection wavelength of 330 nm with a column temperature of 30 ° C and a flow rate of 2.5 ml · min -1 with a sample volume of 0.3 ml. The structures of the compounds were identified by MS and NMR. The immunosuppressive activity of the isolated compounds was investigated by CCK-8 method. Results: Five phenylpropanoids were isolated at the same time. They were erythro-guahexylglycerol, (7R, 8S) -syringoylglycerol, (7S, 8S) -syringoylglycerol, 3-methoxyphenyl- O-methoxycinnamaldehyde. CCK-8 method demonstrated that the inhibitory rates of compound 3 on T cells and B cells at a concentration of 800 μmol·L-1 were both above 20%, showing a certain immunosuppressive effect. CONCLUSION: The method for the separation of phenylpropanoid is simple and has good separation effect. Five phenylpropanoids can be prepared simultaneously. Among them, compound 3 has some immunosuppressive activities.
