目的:观察清脉饮及其拆方对兔动脉粥样硬化闭塞症eNOS、脂联素表达的影响。方法:将42只SPF级雄性白兔随机分为空白组、模型组、全方组、软坚组、活血组、清法组、西药组各6只,实验组建立动脉粥样硬化闭塞症动物模型,于术后8周取材。采用逆转录-聚合酶链反应技术检测eNOS mRNA、脂联素mRNA的活性表达。结果:与空白组比较,模型组、全方组、软坚组、活血组白兔的eNOS mRNA及脂联素mRNA的表达均有所降低(P<0.05或P<0.01),清法组、西药组白兔的eNOS mRNA及脂联素mRNA的表达均明显增高(P<0.01);与模型组比较,全方组、软坚组、活血组、清法组、西药组白兔的eNOS mRNA及脂联素mRNA的表达均明显增高(P<0.01);清法组、西药组白兔的eNOS mRNA及脂联素mRNA的表达均明显高于全方组、软坚组、活血组(P<0.01)。结论:清脉饮及其拆方可有效促进eNOS mRNA、脂联素mRNA表达,从而抑制氧化应激,减轻内皮细胞损伤,达到抗ASO的效果,以清法组为佳。
Objective: To observe the effects of Qingmai Decoction and its decomposed formulas on the expression of eNOS and adiponectin in rabbits with atherosclerosis obliterans. Methods: Forty-two SPF male rabbits were randomly divided into six groups: blank control group, model group, all prescription group, Ruanjian group, Huoxue group, Qingfa group and western medicine group. Animal models of atherosclerosis obliterans , Drawn at 8 weeks after surgery. The expression of eNOS mRNA and adiponectin mRNA was detected by reverse transcription-polymerase chain reaction (RT-PCR). Results: Compared with the blank group, eNOS mRNA and adiponectin mRNA expression in the model group, the whole group, the soft solid group and the activating blood group were decreased (P <0.05 or P <0.01) Compared with model group, eNOS mRNA and adiponectin mRNA expression in group B were significantly increased (P <0.01). Compared with model group, eNOS mRNA and lipid in whole group, Ruanjian group, Huoxue group, Qingfa group and western medicine group The expression of eNOS mRNA and adiponectin in rabbits in Qingcao and western medicine groups were significantly higher than that in Quanfang, Ruanjian and Huoxue groups (P <0.01) . Conclusion: Qingmai Decoction and its decomposed formulas can effectively promote eNOS mRNA and adiponectin mRNA expression, thereby inhibiting oxidative stress, reducing endothelial cell injury and achieving anti-ASO effect.
