以阿克苏骏枣为原料,采用生物复合酶解对骏枣多糖进行提取,以多糖得率和纯度为指标,探讨酶解温度、酶解时间、酶解pH、加酶量对多糖得率和纯度的影响,通过响应面分析法优化骏枣多糖最佳提取工艺,并探讨骏枣多糖的聚酰胺柱层析脱色工艺条件。确定骏枣多糖最佳的生物复合酶解提取工艺条件为:酶解温度60℃,酶解时间65 min,pH6,加酶量1%。在此条件下,多糖得率为12.29%,纯度为39.23%。聚酰胺柱层析最佳脱色工艺为:聚酰胺用量为骏枣粗多糖的4倍,用1倍柱体积的去离子水以1.5 m L/min的流速进行洗脱。
