目的 :探讨 2型糖尿病 (T2DM )患者外周血淋巴细胞 (PBL)凋亡与一氧化氮 (NO)、血糖浓度的关系。方法 :用琼脂糖凝胶电泳法测定 5 3例T2DM [DM组 ,其中无血管并发症 (DMI)组 2 6例 ,伴血管并发症 (DM 2 )组 2 7例 ,]患者及 48例正常人 (NC组 )PBL在体外培养 2 4小时的凋亡发生率 ,同时观察血清NO、葡萄糖浓度及糖化血红蛋白 (HbA1c)的变化。结果 :DM组细胞凋亡、NO、葡萄糖、HbA1c水平均明显高于NC组 (P <0 .0 5 ) ,此现象DM 2组较DMI组更明显 (P<0 .0 5 )。直线相关分析显示 ,细胞凋亡分别与NO、BG、HbA1c及NO分别与餐后 2小时血糖 (PBG2h)、HbA1c呈显著正相关。结论 :2型糖尿病患者外周血细胞凋亡的增强与糖尿病代谢紊乱以及其慢性血管并发症的发生发展有着密切的关系。
Objective: To investigate the relationship between the apoptosis of peripheral blood lymphocytes (PBL) and the levels of nitric oxide (NO) and blood glucose in patients with type 2 diabetes mellitus (T2DM). Methods: 53 patients with T2DM [DM group (26 in DMI group, 27 in DM 2 group) and 48 normal controls were determined by agarose gel electrophoresis Human NC group PBL cultured in vitro for 24 hours, the incidence of apoptosis, while observing serum NO, glucose concentrations and changes in HbA1c. Results: The levels of apoptosis, NO, glucose and HbA1c in DM group were significantly higher than those in NC group (P <0.05). This phenomenon was more obvious in DM 2 group than in DMI group (P <0.05). Linear correlation analysis showed that there was a significant positive correlation between apoptosis and NO, BG, HbA1c and NO respectively with two hours postprandial blood glucose (PBG2h) and HbA1c. Conclusion: The increase of peripheral blood cell apoptosis in patients with type 2 diabetes is closely related to the disorder of diabetes mellitus and the occurrence and development of chronic vascular complications.
