目的:研究氢化可的松、氟美松、羟基脲三种免疫抑制剂对白细胞吞噬活性的直接影响:Zn2+、Mg2+Ge-132对氢化可的松的拮抗作用。方法:采用改良的人全血化学发光法测定,①三种免疫抑制剂按不同剂量加入测定血中时白细胞的化学发光值,②Ge-132中的Ge及Zn2+、Mg2+相当于血浆浓度中Ge、Mg、Zn元素含量的10倍加5μg氢化可的松时白细胞化学发光值。结果:①氢化可的松、羟基脲对白细胞吞噬活性具有直接抑制作用,而氟美松对白细胞吞噬活性在低浓度时具有增强及在高浓度时具有抑制的双效作用;②Zn2+使氢化可的松对白细胞吞噬活性的抑制作用加强,而Mg2+具有拮抗氢化可的松的抑制作用,有机锗(Ge-132)不影响氢化可的松的抑制作用。结论:①为三种免疫抑制剂量大小与抑制白细胞吞噬功能提供定量关系参考,②为金属元素对免疫抑制剂的拮抗作用提供参考依据
AIM: To investigate the direct effects of three immunosuppressants, hydrocortisone, flumethasone and hydroxyurea, on leukocyte phagocytosis: antagonistic effects of Zn2 + and Mg2 + Ge-132 on hydrocortisone. Methods: The chemiluminescence method of human whole blood was used to measure the chemiluminescence of leukocytes when the three immunosuppressive agents were added into the blood at different dosages. Ge and Zn2 +, Mg2 + in Ge-132 corresponded to Ge in plasma, Mg, Zn element content of 10 times plus 5μg hydrocortisone when the value of the chemiluminescence. Results: (1) Hydrocortisone and hydroxyurea had a direct inhibitory effect on leukocyte phagocytosis, while the phagocytosis activity of levometasone was enhanced at low concentration and inhibited at high concentration. (2) Zn2 + The inhibition of phagocytosis of leukocytes was enhanced by pine, while Mg2 + antagonized the inhibition of hydrocortisone. Ge-132 did not affect the inhibition of hydrocortisone. Conclusions: ①To provide a quantitative reference for the size of three immunosuppressive drugs and their inhibition of leukocyte phagocytosis, ② to provide a reference for the antagonism of metal elements on immunosuppressive drugs
