目的:研究葡萄籽提取物(GSE)对雄激素依赖性前列腺癌LNCaP细胞和雄激素非依赖性前列腺癌PC-3细胞血管内皮生长因子(VEGF)和三种性激素睾酮(T)、孕酮(P)、雌二醇(E_2)的影响。方法:实验分为四组:A_1组:LNPCa细胞应用正常细胞培养液培养;A_2组:LNCaP细胞应用含30μg/ml GSE的培养液培养;B_1组:PC-3细胞应用正常细胞培养液培养;B_2组:PC-3细胞应用含300μg/ml GSE的培养液培养。常规培养48 h后收集四组细胞的上清液,用定量酶联检测法和化学发光酶免疫检测法检测VEGF和三种性激素T、P、E_2的浓度变化。结果:与A_1组相比,A_2组VEGF含量降低(P<0.05),T含量升高(P<0.05),E_2含量无明显变化(P>0.05),P未检测到;与B_1组相比,B_2组VEGF含量显著降低(P<0.01),E_2含量升高(P<0.05),T和P含量无明显变化(P>0.05)。结论:GSE在体外能降低前列腺癌LNCaP与PC—3细胞VEGF的含量,提示其可能具有抑制前列腺癌侵袭转移的作用。
Objective: To study the effect of grape seed extract (GSE) on the expression of vascular endothelial growth factor (VEGF) and three sex hormones testosterone (T) and progesterone (P) in androgen-dependent prostate cancer LNCaP cells and androgen independent prostate cancer PC- ), Estradiol (E_2). Methods: The experiment was divided into four groups: A_1 group: LNPCa cells cultured in normal cell culture medium; A_2 group: LNCaP cells cultured in medium containing 30μg / ml GSE; B_1 group: PC-3 cells cultured in normal cell culture medium; Group B_2: PC-3 cells were cultured in a medium containing 300 μg / ml GSE. After 48 hours of routine culture, the supernatant of the four groups of cells was collected. The concentrations of VEGF, three sex hormones T, P and E 2 were detected by quantitative enzyme-linked immunosorbent assay and chemiluminescent enzyme immunoassay. Results: Compared with A_1 group, the content of VEGF in A_2 group was lower (P <0.05), the content of T was higher (P <0.05) and the content of E_2 was not changed (P> 0.05) (P <0.01). The content of E 2 increased (P <0.05) and the contents of T and P in B 2 group had no significant change (P> 0.05). CONCLUSIONS: GSE can decrease the content of VEGF in LNCaP and PC-3 cells in vitro, suggesting that GSE might inhibit the invasion and metastasis of prostate cancer.
