试验目的在于研究土壤和活性污泥的最适DNA提取方法。采用9种不同的提取方法,对所提取DNA的浓度、纯度、提取率、片段大小、16SrDNA PCR结果进行了比较,结果表明,在土壤中,液氮研磨法提取DNA最优:DNA质量浓度为800mg/L,OD260/OD280为1.7。提取率为320μg/g;在活性污泥中,液氮浸泡法提取DNA最优:DNA质量浓度为1 485mg/L,OD260/OD280为1.59,提取率为594μg/g。
The purpose of the experiment was to study the optimum DNA extraction method for soil and activated sludge. Nine different extraction methods were used to compare the concentration, purity, extraction rate, fragment size and 16S rDNA PCR results of the extracted DNA. The results showed that DNA was the best method for DNA extraction by liquid nitrogen grinding method. The DNA concentration was 800mg / L, OD260 / OD280 of 1.7. The extraction rate was 320μg / g. In the activated sludge, the optimum conditions for DNA extraction were liquid nitrogen immersion method: DNA concentration of 1 485mg / L, OD260 / OD280 of 1.59 and extraction rate of 594μg / g.
