目的 研究鼻咽癌 p5 3与 bcl- 2基因表达的相互关系。方法 对 38例鼻咽癌和 16例非癌样本中 p5 3和 bcl- 2 m RNA进行逆转录聚合酶链反应定量分析及单链构象多态性分析。结果 (1)鼻咽癌组p5 3m RNA的水平较高 ,差异有显著性 (t=2 .14,P<0 .0 5 )。 bcl- 2 m RNA水平亦较高 ,单侧 t检验有显著性。 (2 )非癌组 p5 3m RNA随 bcl- 2 m RNA水平的升高而升高 ,相关具有高度显著性 (r=0 .819,P<0 .0 0 1)。在鼻咽癌组虽然也观察到类似的相关关系 ,但 r值比非癌组小 (r=0 .46 6 ,P<0 .0 0 5 )。 (3)鼻咽癌组及非癌组 p5 3- bcl- 2回归系数 b的 t检验均有显著性。 (4 )逆转录聚合酶链反应 -单链构象多态性分析未发现所检查的基因片段有点突变。结论 鼻咽癌中 ,p5 3和 bcl- 2的表达均增高 ,p5 3的失活可能是导致bcl- 2 m RNA过度表达的原因。
Objective To study the relationship between p5 3 and bcl-2 gene expression in nasopharyngeal carcinoma. Methods Quantitative analysis of p5 3 and bcl-2 mRNA in 38 cases of nasopharyngeal carcinoma and 16 cases of non-cancerous samples was performed by RT-PCR and single-strand conformation polymorphism analysis. Results (1) The level of p5 3m RNA in nasopharyngeal carcinoma was higher (t = 2.14, P <0.05). bcl-2 m RNA level is also higher, unilateral t test was significant. (2) The expression of p5mRNA in non-cancer group increased with the increase of bcl-2mRNA, which was highly significant (r = 0.889, P <0.001). In the nasopharyngeal carcinoma group, although a similar correlation was also observed, the r value was smaller than that in the non-cancerous group (r = 0.46 6, P <0.05). (3) The t-test of p5 3- bcl-2 regression coefficient b in nasopharyngeal carcinoma group and non-cancer group was significant. (4) RT-PCR - single strand conformation polymorphism analysis did not find a little mutation in the gene fragment examined. Conclusion The expressions of p5 3 and bcl-2 are increased in NPC, and the inactivation of p5 3 may be responsible for the overexpression of bcl-2 mRNA.
