目的研究鲨鱼软骨粉提取物对大鼠肺微血管内皮细胞的影响。方法用流式细胞仪(PI染色)测定体外培养的肺微血管内皮细胞中细胞周期和细胞凋亡的变化。结果体外培养的微血管内皮细胞与鲨鱼软骨粉全成份(6mg/ml)、成份A(0.01mg/ml)、成份B(0.01mg/ml)一起培养24h后经流式细胞仪检测发现 ,成份B使细胞周期中G2-M期细胞明显减少(P<0.05) ,成份A使凋亡细胞明显增加(P<0.01)。结论成份B可能是鲨鱼软骨粉中的一种水溶性抑制成份 ,它对微血管内皮细胞的增殖起抑制作用 ,成份A对细胞凋亡的作用 ,还有待于进一步研究
Objective To study the effect of shark cartilage powder extract on rat pulmonary microvascular endothelial cells. Methods The changes of cell cycle and apoptosis in cultured pulmonary microvascular endothelial cells were measured by flow cytometry (PI staining). Results The in vitro cultured microvascular endothelial cells were cultured with shark cartilage powder (6 mg/ml), component A (0.01 mg/ml), and component B (0.01 mg/ml) for 24 h and detected by flow cytometry. G2-M phase cells in the cell cycle significantly reduced (P <0.05), component A significantly increased apoptotic cells (P <0.01). Conclusion Component B may be a water-soluble inhibitor in shark cartilage powder. It inhibits the proliferation of microvascular endothelial cells. The effect of component A on cell apoptosis needs further study.
