目的:研究中药组分体外清除多环芳烃、亚硝胺和自由基等烟气有害物质以及降低烟气细胞毒性的作用,为中药组分用于减少烟气危害发挥保健作用提供研究依据。方法:采用分光光度法检测中药组分对DPPH自由基、超氧阴离子和羟自由基的清除作用;以荧光光谱法测定各组分对多环芳烃的清除作用;以紫外分光光度法测定各组分对挥发性亚硝胺形成的抑制作用;并与抗坏血酸的作用进行比较;采用MTT法检测中药组分影响烟气造成的BEP2D细胞毒性作用。结果:所有中药组分均表现出一定的清除上述多种有害物质的作用,其综合清除能力以银杏叶提取物、甘草总黄酮、原花青素、麦冬总皂苷、淫羊藿总黄酮和黄芪总苷为好。以上6种中药组分可显著降低烟气造成的BEP2D细胞的细胞毒性,表现为细胞活存率(SF)显著上升,细胞平均致死剂量(DML)显著增加。结论:上述中药组分有良好的清除多环芳烃、亚硝胺和自由基等烟气有害致癌物质的作用,在减少烟气对人群的危害和预防烟草引起的各种疾病的发生中可发挥重要保健作用。
OBJECTIVE: To study the role of traditional Chinese medicine components in removing harmful substances such as polycyclic aromatic hydrocarbons (PAHs), nitrosamines and free radicals in vitro and to reduce cytotoxicity of flue gas, and to provide a research basis for traditional Chinese medicine components to reduce the harm of flue gas and exert their health benefits. Methods: The scavenging effects of Chinese herbs on DPPH free radicals, superoxide anion and hydroxyl radical were detected by spectrophotometry. The scavenging effects of each component on PAHs were determined by fluorescence spectroscopy. Points on the formation of volatile nitrosamines inhibition; and compared with the role of ascorbic acid; using MTT assay of Chinese medicine components affect the smoke caused by BEP2D cytotoxicity. Results: All the Chinese medicine components showed a certain amount of removal of the above-mentioned a variety of harmful substances, its comprehensive removal ability to Ginkgo biloba extract, total flavonoids of licorice, proanthocyanidins, Radix Ophiopogon japonicus total flavonoids and total astragalosides As well The above 6 kinds of traditional Chinese medicine components can significantly reduce the cytotoxicity of BEP2D cells caused by flue gas, showing a significant increase of cell viability (SF) and a significant increase of cell lethal dose (DML). Conclusion: The above traditional Chinese medicine components have a good function of eliminating harmful carcinogens such as polycyclic aromatic hydrocarbons (PAHs), nitrosamines and free radicals, and can play a role in reducing the harm of flue gas to the population and preventing the occurrence of various diseases caused by tobacco Important health effects.
