目的:探讨2种海藻溴酚化合物对3株肿瘤细胞Hela,MGC,BGC-823的体外抗肿瘤作用及其作用机制。方法:采用MTT比色法检测不同浓度溴酚化合物对肿瘤细胞的抑制作用;用流式细胞术分析2种化合物对3种肿瘤细胞的细胞周期和非整倍体的诱导作用。结果:2种海藻溴酚化合物对肿瘤细胞均有较强的抑制作用,其中对Hela细胞的抑制活性最强。2种海藻溴酚化合物均不能引起细胞凋亡,但能使肿瘤细胞的倍性改变,产生非整倍体,并使细胞周期受到影响。对Hela细胞和MGC细胞,低浓度能引起亚倍性的产生,较高浓度则引起G1期抑制。对BGC-823细胞,不同浓度处理均能引起G1期抑制,其抑制程度具有剂量效应。结论:海藻溴酚化合物对肿瘤细胞具有不同的生长抑制活性,其作用机制与诱导产生非整倍体和细胞周期抑制有关。
Objective: To investigate the anti-tumor effect and mechanism of two kinds of seaweed bromophenols on three tumor cells Hela, MGC and BGC-823 in vitro. Methods: MTT colorimetric assay of different concentrations of bromophenol compounds on tumor cells inhibition; flow cytometry analysis of two compounds on the three kinds of tumor cell cycle and aneuploidy induction. Results: The two kinds of alginate bromophenol compounds had a strong inhibitory effect on tumor cells, of which Hela cells had the strongest inhibitory activity. The two kinds of alginate bromophenol compounds can not cause apoptosis, but can make the ploidy of tumor cells change, resulting in aneuploidy, and the cell cycle is affected. In Hela cells and MGC cells, low concentrations induced the production of hypopyx and higher concentrations caused G1 inhibition. In BGC-823 cells, different concentrations of treatment can cause G1 phase inhibition, the degree of inhibition with a dose-effect. Conclusion: The alginate bromophenol compound has different growth inhibitory activity on tumor cells, and its mechanism is related to induction of aneuploidy and inhibition of cell cycle.
