目的:研究当归芍药散对慢性应激抑郁模型大鼠行为及中枢单胺类递质的影响。方法:应用分养和长期不可预见性温和应激造成大鼠抑郁模型,并观察当归芍药散对该模型大鼠的治疗作用。定期测定各组大鼠糖水嗜好程度;敞箱实验方法测定大鼠水平及垂直活动;RP-HPLC-荧光法测定动物大脑额叶皮质中神经递质的含量。结果:与正常对照组比较,模型组大鼠糖水消耗、垂直活动、水平运动显著减少,大脑额叶皮质中NE,DA,5-HT,5-HIAA的含量均明显降低;当归芍药散高剂量可明显升高DA,5-HT,5-HIAA的含量(P<0.01),低剂量仅对5-HIAA含量有明显增高(P<0.01),对NE含量无明显改变。结论:初步表明当归芍药散具有抗抑郁作用,机制可能与调节脑内单胺类神经递质DA,5-HT,5-HIAA的含量有关。
Objective: To study the effect of Danggui Shaoyao San on behavior and central monoamine neurotransmitter in chronic stress depression model rats. Methods: The model of depression was induced by moderate and long-term unpredictable mild stress in rats. The therapeutic effects of Danggui Shaoyao San on the model rats were observed. The levels of glucose and water preference of rats in each group were determined regularly. The open-field test was used to measure the horizontal and vertical activity of rats. The contents of neurotransmitters in the frontal cortex of the brain were measured by RP-HPLC. Results: Compared with the normal control group, the contents of NE, DA, 5-HT and 5-HIAA in the frontal cortex of rats in the model group were significantly lower than those in the normal control group The contents of DA, 5-HT and 5-HIAA were significantly increased (P <0.01). The content of 5-HIAA in low dose group was significantly increased (P <0.01), while the content of NE was unchanged. CONCLUSION: The preliminary results show that Danggui Shaoyao San has antidepressant effect, which may be related to the regulation of brain monoamine neurotransmitters DA, 5-HT and 5-HIAA.
